【Abstract】Objective To investigate the influence of the different dialysis membrane on the interleukin-8 expression in uremic patients. Methods The plasma concentration of IL-8 and the IL-8 production by the cultivated PBMC were measured with enzyme-linked immunoabsorbent assay(ELISA). The gene expression of IL-8 was measured with reverse transcription-polymerase chain reaction(RT-PCR). Results The IL-8 plasma and mRNA level of the patients using the regenerated cellulose membrane (cuprophan CU, cuprammonium rayon CR) is higher than that of those using polysulfone (PS) membrane. The IL-8 plasma level and mRNA expression increases after hemodialysis. The production of IL-8 from PBMC of the patients using CU membrane was more than that of those using PS membrane. In PS group the IL-8 production of PBMC increased after dialysis. But there was no obvious difference of IL-8 production from PBMC between pre-dialysis and post-dialysis in CU group. Conclusion The change of IL-8 mRNA expression and plasma level through the hemodialysis in the regenerated cellulose membrane (especially CU) groups is more significant than that in PS group. The measurement of IL-8 expression is useful to evaluate the biocompatibility of dialysis membranes.
