Abstract

Objective To investigate the effect of heparin on inhibition of cell proliferation as well as on expression of interleukin-1(IL-1) in RPMC (rat peritoneal mesothelial cells) by high glucose, so as to provide some theoretic basis for the clinical use of heparin during continuous ambulatory peritoneal dialysis (CAPD). Methods RPMC were cultured in vitro and MTT method was processed to observe the effects of both high glucose (3.86%) and heparin (1.25U/ml) on cell proliferation. In addition, ELISA analysis were performed to investigate the change of IL-1 in the culture muli. ResuIts High glucose of 3.86% could significantly inhibit cell proliferation as well as induce the expression of IL-1β in RPMC (P<0.01), which could be partially reversed by heparin of 1.25U/ml (P<0.01). Conclusion Heparin could reverse both inhibition of cell proliferation and induction of pro-inflammatory cytokine IL-1 mediated by high glucose in peritoneal mesothelial cells, and delay the occurrence of CAPD associated peritoneal fibrinosis and being beneficial to the process of CAPD.
