肝素、高糖对人腹膜间皮细胞生长及合成细胞外基质的影响

摘要: 【摘要】目的 探讨肝素、高糖对体外培养的人腹膜间皮细胞(HMC)生长和合成细胞外基质影响。方法 3 H-胸腺嘧啶核苷掺入法检测细胞增生;ELISA法检测细胞培养上清纤维连结蛋白(FN)、Ⅰ型胶原(COLⅠ)含量,细胞计数校正其浓度;RT-PCR检测FN基因表达。结果 1.36%葡萄糖介质中培养72h可见HMC 3H-TdR掺入量明显降低(P<0.05),呈时间和剂量依赖关系,高糖环境下肝素对HMC增殖率无明显影响;3.86%葡萄糖可明显促进FN和COLⅠ的分泌,肝素可明显降低高糖环境下FN和COLⅠ的分泌。结论 高糖可能是长期腹透致腹膜硬化的一个重要因素;肝素对保护腹膜功能可能有一定的作用。

Abstract:

【Abstract】Objective To study the effect of heparin and high concentration glucose on cell growth and matrix biosynthesis in cultured human peritoneal mesothelial cells. Methods 3 H-thymidine incorporation for mesurement of cell growth and ELISA kits for detection of fibronectin and collagenⅠin supernatant. Semiquantitative reverse transcription polymerase chain reaction(RT-PCR) for detection of FN. mRNA Results Incubated in 1.36% glucose for 72 hours, HMC 3 H-thymidine incorporation rate decreases significantly(P<0.05). High concentration glucose cause time and concentration dependent decrease in 3 H-thymidine incorporation rate and heparin dosen't infect 3 H-thymidine incorporation rate when HMC was already incubated in high concentration glucose; high concentration glucose can increase the excretion of fibronectin and collagenⅠand heparin can inhibite the increase of fibronectin and collagenⅠexcretion stimulated by it. Conclusion High concentration glucose may play an important role in peritoneal sclerosis during peritoneal dialysis; heparin may be beneficial for protecting the function of peritoneum during peritoneal dialysis.
