成年动物肝细胞基本培养法筛选暨胆红素代谢实验

摘要: 目的 探讨成年动物肝细胞 (adult animal hepatocytes,AAH)基本培养法和胆红素代谢、尿素氮生成的生物活性,寻找体外生物人工肝支持系统(extracorporeal bioartificial liver support system, EBLSS)更理想的生物材料。方法 ①三种基本成分:合成培养基、血清、胰岛素对成年小白鼠肝细胞作15种组合培养观察最长存活期;② 用重肝高胆红素血清培养AAH,观察胆红素代谢、尿素氮生成活性。结果 含20%同种异体雄性小白鼠灭活血清、10U/ml胰岛素的高糖型DMEM培养基作高密度培养时,第14天时仍有20%的存活率;AAH 3h使总胆红素、直接胆红素、间接胆红素各下降 28.88%、21.13%、33.63%。结论 本模式可作为AAH的基本培养法;AAH可成为EBLSS更理想的生物材料。

Abstract: Objective We probed the basic culture methods of adult animal hepatocytes(AAH) which may become more ideal biological material of extracorporeal bioartificial liver support system (EBLSS) and their biological activity of metabolism of bilirubin and synthesis of carbamide. Methods ①Using three kinds of basic components (synthetic medium, serum and insulin), the 15 kinds of combination of cell-cultured methods on adult mice hepatocytes were observed. ② AAH were cultured with hyperbilirubin serum of human severe hepatitis and their biological activities of metabolism of bilirubin and synthesis of carbamide were observed. Results Cells cultured in high metabolism density culture medium and containing 20% inactivated allograph male rat serum,10U/ml insulin, high sugar DMEM still remained 20% survival rate in the 14th day. The metabolic activity of 24 hours-pre-cultured monolayer hepatocytes was the most powerest on bilirubin. The Serum total bilirubin, direct bilirubin and indirect bilirubin fell 28.88%, 21.13%, 33.63% respectively after 3 hours culturing by hyperbilirubin serum of human severe hepatitis. Conclusion This model system can be used as the basic culture medium of adult animal hepatocytes. AAH would become more ideal biological material of EBLSS to substitute the newly born animal hepatocytes and embryo hepatocytes.
