一种简便的正常大鼠腹膜透析模型建立

摘要:

目的 建立1种简便的正常大鼠腹膜透析动物模型。方法 应用SD大鼠,实验分为4组:正常 组、假手术组、4.25%腹膜透析组、1.25%腹膜透析组。自制透析管是经腹部植入后从背部引出,腹膜透析大鼠每只每日透析30ml,腹腔保留1h。在腹膜透析的第1周和第四周检测肾功能和白蛋白,同时观察体重、腹膜透析流出液量及并发症。结果 两组腹膜透析组间白蛋白及体重测定相比无统计学差异,但两者与正常组相比差异有显著性;在第1周及第4周流出液量相似,无统计学差异,腹膜透析的主要并发症为腹膜炎、大网膜包裹及导管移位等。
结论 本实验建立的这种正常大鼠腹膜透析动物模型可模拟人类腹膜透析。

Abstract:

Objective To develop an improved model of peritoneal dialysis in animal that mimic human peritoneal dialysis. Methods The study was done in male SD rats, animals were divided into four groups: normal rats (group 1) and sham operated rats (group 2) and dialysized rats exposed to PD solution4.25% (group 3) and 1.5% (group 4) respectively. The catheters implanted in dialysis rats were tunneled subcutaneously to the back and exchanged PD solutions 30ml per rat without anesthesia in two hours a day. At the first week and the fourth week of the study, the samples of blood were taken from these rats in each group to test serum creatinine and urea nitrogen and albumin, meantime, the body weigh and the mount of exchanged PD solutions were examined. Results All rats underwent PD could be administrated to exchanged PD solution repeatedly, the complications were peritonitis, catheter obstruction and /or dislocation, the rats that could not undergo PD were replanted the catheter to continue PD. The exchanged solution in the first week and the fourth week were similar between group 3 and group 4. Conclusion The conscious rat model for PD does allow regular fluid drainage, and it is a drainage model that mimiced human peritoneal dialysis.
